Why are we demanding Iplex for ALS patients?
Iplex is a drug based on the naturally occurring hormone Insulin Growth Factor-1
(IGF-1). In the human body, 80% of IGf-1 is bound to a very specific protein:
IGF Binding Protein-3 (IGFBP-3). Insmed is the only company in the world capable of
and actively producing the combination of these two proteins. IGFBP-3 has been
scientifically proven to deliver a greater dose of IGF-1 and to extend the half-life by
several hours over free-IGF-1, allowing for a once-a-day treatment. This extension of
half-life also allows the IGF-1 to reach its intended targets, we presume, crossing the
blood-brain-barrier,where it has the greatest opportunity to protect neurons, promote
new growth and fix damaged cells for ALS. It is FDA approved for the treatment of
Severe Short Stature.

Iplex is currently in a Phase-2b national placebo-based trial for the treatment of
Myotonic Dystrophy, which will be complete by December 2008. The results of the
Myotonic Dystophy ph2a trial showed that Iplex improved lean muscle mass, improved
gastrointestinal function, improved walking endurance and cognitive function, while
relieving pain associated with the condition.

Italian ALS patients, who have been receiving Iplex for one-and-a-half years now,
have reported significant improvements fromthe use of Iplex for ALS, as reported in
various media outlets and court proceedings, both for new patients and patients
seeking extension for the continued use of Iplex in the Expanded Access Program
conducted by Insmed, with a partnership of Cephalon and the Italian Ministry of Health.
Several Americans, who had brief access to Iplex, also reported rapid improvements.
ALS patients around the world deserve to have an opportunity to try Iplex for
themselves and discover if it might accomplish what no other treatment to date has
ever done: substantially slow down the progression of ALS and possibly even restore
lost motor function.


Warum fordern wir Iplex für ALS-Patienten?
Iplex ist ein Medikament, das auf dem natürlich vorkommenden, insulinähnlichen
Wachstumsfaktor IGF-1 basiert. Im menschlichen Körper ist IGF-1 mit dem bindenden
Protein IGF-3 (IGFBP-3) verbunden. Insmed ist die einzige Firma weltweit, die in der
Lage ist, eine Kombination aus diesen beiden Medikamenten herzustellen. Es ist
wissenschaftlich erwiesen, dass IGFBP-3 IGF-1 wesentlich länger im Körper bewahrt
als freier IGF-1, so dass eine einmalige Behandlung am Tag ausreichend ist. Dieser
längere Verbleib im Körper ermöglicht auch, dass IGF-1 die beabsichtigen Ziele im
Körper erreicht, indem es anscheinend die Blut-Hirn-Schranke überwindet und somit
die größtmögliche Chance hat, bei ALS-Patienten Neuronen zu schützen, neues
Wachstum zu fördern und beschädigte Zellen zu reparieren. Iplex wurde in den USA
bereits von der Arzneimittelzulassungsbehörde FDA zur Behandlung von Kleinwüchsig-
keit zugelassen.

In den USA befindet sich Iplex gerade in Phase 2 einer Placebo-kontrollierten Studie
zur Behandlung der Myotonen Dystrophie, die im Dezember 2008 beendet werden soll.
Die Ergebnisse bisher zeigen, dass Iplex schwache Muskeln stärkt, die Magen-Darm-
Funktion, die Ausdauer beim Gehen sowie kognitive Funktionen verbessert, während
es auch die Schmerzen, die mit dieser Krankheit verbunden sind, lindert.

Italienische ALS-Patienten, die Iplex seit eineinhalb Jahren bekommen, berichten eben-
falls über erhebliche Verbesserungen ihres Zustands. Weitere Patienten versuchen,
in das in Italien existierende ‚Erweiterte Zugangsprogramm’ zu Iplex zu gelangen, das
von Insmed in Zusammenarbeit mit Cephalon und dem italienischen Gesundheits-
ministerium durchgeführt wird. Einige US-Amerikaner, die kurzzeitig Zugang zu Iplex
hatten, berichten ebenfalls schnelle Verbesserung ihres Zustandes. Als-Patienten in
der ganzen Welt verdienen die Möglichkeit, Iplex selbst zu bekommen und zu sehen, ob
es das erreicht, was bisher keine andere Behandlung erreicht hat: die substantielle
Verlangsamung der Progression von ALS und möglicherweise sogar die Wiederherstell-
ung von bereits verloren gegangenen motorischen Funktionen.

ALS Patients Protest Legal Dispute Preventing Access to

Life-Saving Drug

GENENTECH-TERCICA*-INSMED: RELEASE IPLEX NOW!

Support patients with Amyotrophic Lateral Sclerosis (Lou Gehrig’s disease) now by joining

our demonstration to force Genentech, Tercica and Insmed to resolve their legal dispute and

release IPLEX to provide symptom relief for this horrible disease.  We are calling on all

persons with ALS, caregivers, friends, family, media and others to join us in protesting this

devastating travesty of justice.

IPLEX is a biomedical pharmaceutical that reduces the symptoms for ALS patients and

provides significant relief and improvements, a previously unheard-of achievement although

not specifically developed for that purpose.  Despite that, on March 7, 2007, a settlement

agreement between Genentech, Tercica and Insmed removed IPLEX from ALS patients in the

US and throughout the world, with the exception of Italian ALS patients, a cruel settlement

exclusion.While Italian ALS patients benefit from IPLEX, the rest of the ALS world suffers

debilitation, deprivation and death.

IPLEX is an improved version of Increlex, a drug originally intended to improve severe short

growth stature. While both drugs accomplish that to a greater or lesser degree, only IPLEX

significantly improves the quality of life for ALS patients.  Despite that, persons with ALS lost

access to IPLEX and are offered Increlex.  After many months of usage, it is clear that IPLEX

is the only drug that offers the greatest possible benefit to ALS patients.  Nevertheless, it remains

unavailable due to the greed of Genentech, Tercica and Insmed, all of whom agreed to this

immoral settlement agreement in the name of corporate and personal profit.

Attempts to procure IPLEX have failed, despite continuous contact with the CEO’s of all three

corporations, media,  judiciary, attorneys, Senators, Representatives and other government

officials.  Each blames or points to the other.  No one takes responsibility for this travesty. 

Public demonstration is now the only recourse left to the ALS community.

ALS robs its victims of all voluntary muscles, sentencing its victims to what is called “a brief life

in a glass coffin” and death within 2 to 4 years.  IPLEX is the only drug yet developed that

significantly slows and in some cases reverses this devastating disease. 

Our hope is this demonstration will alert the public, our congress and the media to help force

Genentech, Tercica and Insmed to release IPLEX.  Join us in this critical effort. Help make

IPLEX available to ALS patients to improve the quality of their lives, for however long that is.
